OVERVIEW

The course is developed with a theoretical (36 hrs) and practical lessons (60 hrs for each student) concerning the pre-treatment and the quantitative determination of substances of pharmaceutical interest, using classical and innovative methods. The topics of the theoretical course are conducted in close correlation with the practical trainings in laboratory. The frontal teaching can be also carried out remotely in telematic mode.

AIMS AND CONTENT

LEARNING OUTCOMES

The course consists of theoretical lessons and laboratory activities and aims to give the basic notions for the quantitative determination of compounds with medicinal use. In particular, during the lab activities, quantitative assays of organic and inorganic compounds are carried out.

AIMS AND LEARNING OUTCOMES

The course aims to give the student the theoretical basis and the practical skills of the main methodologies of sample purification and quantitative analysis in Medicinal Chemistry. At the end of the course, the student should: i) have a deep knowledge of the theory at the basis of lab experiences; ii) carry out the lab procedures with confidence; iii) critically interpret the experimental results.

TEACHING METHODS

The course consists of theoretical lessons and individual or group laboratory activities for a total of 96 hours. Because of the COVID-19 emergency, part of theory lessons could be conducted remotely in telematic mode.

SYLLABUS/CONTENT

Theoretical topics

Guidelines on laboratory safety and chemical use

Introduction to the quantitative determination of compounds registered in FU XII Ed. and FE VIII Ed.

Overview of the analytical methods. Classification and selection of analitycal methods. Chemical analysis stages. Chemical, apparatus and methods for the quantitative determination of compounds with medicinal uses. Calculation used in pharmaceutical analysis. Sampling, standardization and calibration.

Introduction to volumetric titrations in pharmaceutical analysis: chemical equilibria in volumetric analysis. Preparation of standard solutions.

Techniques of preparing sample: filtration, extraction, distillation, chromatography

Quantitative determination and applications: neutralization titrations, complexation and precipitation reactions and titrations, oxidation/reduction titrations, potentiometry.

Atomic absorption spectrometry (AAS) and applications.

Practical trainings

Extraction with solvent, distillation in vacuo. Chromatographyc separation and analysis of a mixture of drugs. Quantitative determination of drugs by mean of neutralization titrations, complexation and precipitation reactions and titrations, oxidation/reduction titrations, potentiometry and AAS.

EXAM DESCRIPTION

The exam consists of an oral test of 30-40 minutes duration. For the final evaluation, the results of the laboratory activities will be considered. Because of the COVID-19 emergency, exams could be conducted remotely in telematic mode.

ASSESSMENT METHODS

Over the course, the student must acquire progressive ownership of the experimental techniques used, learning to independently perform the analysis, interpret the data in the right way and correctly produce a lab report. In the oral test, the theoretical knowledge at the basis of the laboratory activities is evaluated.
