AIMS AND CONTENT

AIMS AND LEARNING OUTCOMES

The objectives of the propaedeutic laboratory are the preparation of the students with a first presentation and exercise on the subjects that will be deepened in the various specialized sectors of laboratory foreseen by the annual internship. The Propaedeutic Laboratory of the second year of the course of Biomedical Laboratory Techniques aims to impart to students some basic notions as well as the performance of some tests in autonomy

SYLLABUS/CONTENT

Introduction to the Electrophoresis and Specific Protein Sector: biological materials and collection; anticoagulants; treatment, conservation and centrifugation; identification of biological samples.

• Electrophoretic session setup: electrophoresis principle and methods; Equipment and accessories (power supply, electrophoretic chamber, buffer, dye, densitometer); Stages of electrophoretic running (application, migration, coloring, detection).

• Electrophoretic separation of proteins: standard electrophoretic profile; Fractions (albumin-α1-α2-β1-β2-γ): Specific proteins found in the electrophoretic tracing.

• IFE (gel) electrophoretic immunofixation

• Bence Jones Protein Research (gel)

• Automatic electrophoresis analyzers

•Instrumental analytical techniques: photometry spectrum, turbidimetry, nephelometry and potentiometry. Principle: Lambert-Beer law, application. Calibrations. Internal and external quality control. Analytical interferences. Technical evaluation of dosages in clinical chemistry.

• Recognition of culture media

• Seeding test on culture media

• Recognition of bacterial colonies on different culture media

• Conduct of some preliminary tests to identify bacterial colonies (oxidase, catalase, etc.)

• Preparation slide for staining

• Execution of Gram staining

• Microscope slide reading with colonies recognition

• Reading and interpretation of manual antibiograms (kirby Bauer, E test)

  • Acceptance of samples in pathological anatomy; Material suitability and fixation based on its type. Which containers to use, which tools and their correct use

• Reduction: how to prepare the bench for reduction. How to perform final cleaning and disinfection. Tutorials on the creation of worksheets and dictation of the macroscopic. Exercises on the use and type of instruments (scalpels, pliers, etc.). How and which biocassettes to use; how to sample all biopsies (the exercises will consist in having the student take samples, for example using material of animal origin).

• Cutting: the Microtome. Description and routine maintenance. Cutting exercises

• Staining (and mounting of slides): which tools; preparation of coloring solutions

• Use of the optical microscope to ascertain the occurred coloring (internal and external quality controls).

• Cytology. Techniques for setting up cytological materials. Use of the cytocentrifuge
