AIMS AND LEARNING OUTCOMES

The objectives of the propaedeutic laboratory are the preparation of the students with a first presentation and exercise on the subjects that will be deepened in the various specialized sectors of laboratory foreseen by the annual internship. The Propaedeutic Laboratory of the third year of the course of Biomedical Laboratory Techniques aims to impart to students some basic notions as well as the performance of some tests in autonomy

SYLLABUS/CONTENT

  • Dosage of acid metabolites of urinary catecholamines: VMA, HVA. 5-HIAA
    • Sample preparation and extraction on DISP. WITH THE
    • Use of HPLC system for chromatographic separation and Electrochemical detector
    • Calibration of the HPLC system and calculation of the calibration factor
    • Calculation of VMA, HVA, 5-HIAA concentration in Controls and samples
    • Main ways of expressing the concentrations of solutes in solutions, of
    • greater use in the laboratory:% weight,% volume, molarity, molality, ppm.
    • How to dilute a 35% HCl solution (concentrated commercial solution for laboratories) to obtain: * HCl * * 6N * solution (concentration used to prepare the preservative for urine samples for Catecholamine, Metanephrine etc. HPLC assays) ); solution * HCl * * 0.05 N * (concentration used to prepare calibrators and controls for Catecholamines, Metanephrines etc. in HPLC)
    • reading and interpretation of IHC anti-data-sheet
    • preparation of protocols for testing an antibody
    • Concentrated antibody dilutions (simulations)
    • Crafts required with IHC tools (similar preparation of an immunohistochemistry session)

    • Overview of the erythrocyte system. antigen-antibody, ABO system, Rh phenotype, Kell system
    • Preparation and execution of a direct and indirect group determination on plate, in test tube, on glass and in ticket; interpretation of results, discussion, resolution of any problems arising from discrepancies in results
    • Determination of direct group and Coombs test directed on funiculus blood, execution (if tcd positive) of the eluate, preparation of the antibody research, identification and interpretation of the results with the master list.
    • Determination of direct / indirect group and indirect Coombs test on maternal blood; in case of positivity, possible preparation of the antibody research, identification and interpretation of the results with the master list.
    • Presentation and discussion of various cases of MEA and MEN
    • Direct / indirect Coombs test on donor / patient samples; in case of positivity, execution of the fractionated Coombs test in the first case and, in the second case, preparation of the panel with 11 cells, which, based on the positivity obtained, can be performed on the ticket or in the test tube.
