AIMS AND CONTENT

AIMS AND LEARNING OUTCOMES

Acquire knowledge and practical skills related to molecular biology techniques, immunohaematological, immunohistochemical, chromatographic and flow cytometric techniques.

TEACHING METHODS

Learning activities of basic laboratory techniques with compulsory attendance of 100% of the planned hours of the laboratories and direct learning through the use of tools with the guidance of professionals belonging to the same Professional Profile. There are no hours of individual study.

 
Verification of the ability to carry out laboratory tests with a description of the methods of carrying out and of the purposes of the test for clinical-diagnostic investigations

 

SYLLABUS/CONTENT

Pathological anatomy sector 140 h:

• Immunohistochemical techniques

• Preparatory treatment of the material for immuno-histochemical examinations

• Basic immunohistochemical techniques (manual, semi-automatic and automatic)

• Main markers in immuno-histochemistry (data-sheet reading, optimal antibody new dilution, etc.)

• Use of positive and negative controls

• Preparation of tampons; tool maintenance and management

• Immuno-histochemistry on cytological material

• Preparation of preparations for extemporaneous diagnostics - Elective stains for intraoperative examinations - cytological preparation for intraoperative examination

• In situ hybridization techniques (FISH –CISH-SISH); manual, semi-automatic, and automatic techniques

• Preparatory treatment of the material for FISH analysis (histological, cytological, decolored, peripheral blood, bone marrow aspirate)

• Main types of probes reading the data sheet and setting up the methods for new reagents

• Use of controls (probe with internal control); tool maintenance and management.

• Microscopic evaluation of the preparations carried out and their adequacy, any changes to be made

 

Molecular biology sector 175 h:

• Organization of areas and behavioral norms in the molecular biology laboratory

• Nucleic acid extraction with manual and automatic methods used in the laboratory

• Dosage and evaluation of nucleic acid quality

• PCR techniques, GEL electrophoresis

• Sequencing

• Interpretation of results

• Maintenance and use of laboratory instruments

• CQI and VEQ quality controls

 

Immunohematology sector 140 h:

• Blood group on adults; Blood group + TCD on newborns

• Execution of phenotype Rh

• Weak “D” research test execution

• Direct / fractionated Coombs test; Indirect Coombs test

• Research and identification of anti-erythrocyte antibodies

• Elution of autoantibodies

• Automation in immunohaematology;

• Role and activities of a TSLB in a SIT

• Emovigilance and security

• Adverse reactions

CYTOFLUORIMETRY OR CHROMATOGRAPHY SECTOR 105 H:

Flow cytometry

• Knowledge of the physical and technological bases of flow cytometry: general scheme of a flow cytometer (fluidics, optics, electronics); significance of the cellular physical parameters evaluated in cytometry; fluorescence signals; description of fluorochromes; clearing operation; meaning of multiparametric analysis; gating strategy; data representation

• Direct and indirect coloring techniques of biological samples

• Main application methodologies for the study of phenotype and lymphocyte functions, such as cytokine production and proliferation

Chromatography

• Demonstration and characteristics of the functioning of the modules of an HPLC

• Extraction of a sample on a resin column

• Use of the S.I. in different methods

• Reading of a chromatogram and possible changes to the retention times of the peaks

• Intervention on the reading program to highlight and recognize the various operations in order to recognize a substance in the analytical phase

• Exercise on the various methods in the elution phase of a sample

• Procedure for stabilizing a column and identifying the various columns on an HPLC module

• Demonstration and operation of an LC-MS / MS; extraction of a sample for analysis in LC-MS / MS

• Evaluation of an analysis calibration curve in LC-MS / MS

The practical internship begins after the end of the lessons of the first semester from December to February and after the lessons of the second semester from April-May to September with a break in August.

Verification of the ability to carry out laboratory tests with a description of the methods of carrying out and of the purposes of the test for clinical-diagnostic investigations.
The exam takes place at the end of the annual internship. The student must demonstrate that he has acquired the expected objectives.
