CODE 70735 ACADEMIC YEAR 2023/2024 CREDITS 1 cfu anno 3 TECNICHE DI LABORATORIO BIOMEDICO 9293 (L/SNT3) - GENOVA SCIENTIFIC DISCIPLINARY SECTOR MED/46 LANGUAGE Italian TEACHING LOCATION GENOVA SEMESTER Annual MODULES Questo insegnamento è un modulo di: PRACTICAL-CLINICAL INTERNSHIP - 3RD YEAR TEACHING MATERIALS AULAWEB AIMS AND CONTENT AIMS AND LEARNING OUTCOMES The educational objectives of the professionalizing laboratory are the introduction and presentation of the laboratory disciplines that the students will have to acquire in the year of enrollment before the start of the curricular internship at the affiliated institutions, divided among the various teachers specialized in the individual disciplines and after classroom lessons related to specific disciplines. The Preparatory Laboratory of the III year of the Biomedical Laboratory Techniques course aims to impart to the students some notions related to the attendance of specialized laboratories as well as the carrying out of some tests independently TEACHING METHODS Practice in the Laboratory with the supervision of Clinical Tutors or Internship Guides SYLLABUS/CONTENT Chromatographic techniques: dosage of the acid metabolites of urinary catecholamines: VMA, HVA. 5-HIAA; sample preparation and extraction on DISP. WITH THE; use of HPLC system for chromatographic separation and electrochemical detector; calibration of the HPLC system and calculation of the calibration factor; calculation of the concentration of VMA, HVA, 5-HIAA in the controls and samples; main ways of expressing the concentrations of solutes in solutions, most commonly used in the laboratory:% weight,% volume, molarity, molality, ppm; how to dilute a 35% HCl solution (concentrated commercial solution for laboratories) to obtain: a) * HCl * * 6N * solution (concentration used to prepare the preservative for urine samples for assays of Catecholamines, Metanephrines etc. in HPLC); b) solution * HCl * * 0.05 N * (concentration used to prepare calibrators and controls for Catecholamines, Metanephrines etc. in HPLC). Molecular biology techniques: Manual skills necessary for the practice of molecular biology techniques. Choice of the appropriate protocol for the diagnostic purpose. Preparation of the necessary material and reagents (simulations). Dilutions of concentrated reagents (simulations). Preparation of the sequencing reaction (simulations). Use of thermal cycler (simulations). Knowledge and operation of automatic sequencer. Reading and interpretation of the electropherogram. Immunohematology techniques: ABO system, Rh phenotype, Kell system; preparation and execution of a direct and indirect group determination on plate, test tube, slide and coupon and interpretation of the results; direct group determination and direct Coombs test on funicular blood; direct / indirect group determination and indirect Coombs test on maternal blood; direct / indirect Coombs test on donor / patient samples in coupons or in test tubes. Immunohistochemical techniques: reading and interpretation of the IHC antibody data sheet; preparing protocols for testing an antibody; Concentrated antibody dilutions (simulations); Manual skills required with IHC tools; facsimile preparation of an immunohistochemistry session; slide preparation and choice of effective IHC slides; knowledge and choice of effective reagents for an IHC session; protocol for testing the antibody reaction and evaluation of positive controls; preparation of TRIS Buffer solution; preparation of the cytological material for IHC tests; definition of immunocytochemical protocols; switching on and checking the functionality of the Cryostat; positioning of fabrics for cutting test (animal origin); Cryostat cutting tests on different fabrics (animal origin): practical problems to be faced; colors that can be performed; slide control.
